Serum levels of soluble B and T lymphocyte attenuator predict overall survival in patients undergoing immune checkpoint inhibitor therapy for solid malignancies

چکیده
Therapy with immune checkpoint inhibitors (ICIs) can lead to durable tumor control in patients various advanced stage malignancies. However, this is not the case for all patients, leading an ongoing search biomarkers predicting response and outcome ICI. The B T lymphocyte attenuator (BTLA) expressed on cells that was shown modulate therapeutic responses. Here, we evaluate circulating levels of its soluble form, (sBTLA), as a biomarker prediction treatment ICI therapy. Serum sBTLA were analyzed by multiplex immunoassay n = 84 receiving therapy solid malignancies 32 healthy controls. BTLA expression evaluated peripheral blood mononuclear subset (n 6) using multicolor flow cytometry. Baseline serum significantly higher cancer compared Importantly, independent prognostic factor overall survival (OS). As such, initial above calculated cutoff value (311.64 pg/mL) had median OS only 138 days 526 below (P .001). Uni- multivariate Cox regression analyses confirmed role context Finally, observed significant correlation between frequency CD3 + CD8 BTLA+ blood. Thus, our data suggest could represent noninvasive predict therapy, helping select eligible candidates.

B and T Lymphocyte Attenuator is a Target of miR-155 during Naive CD4+ T Cell Activation
Background: MicroRNA-155 (miR-155) is upregulated during T cell activation, but the exact mechanisms by which it influences CD4+ T cell activation remain unclear. Objective: To examine whether the B and T lymphocyte attenuator (BTLA) is a target of miR-155 during naïve CD4+ T cell activation. Methods: Firefly luciferase reporter plasmids pEZX-MT01-wild-type-BTLA and pEZX-MT01-mutant-BTLA were ...

B and T lymphocyte attenuator restricts the protective immune response against experimental malaria.
The immune response against the blood stage of malaria has to be tightly regulated to allow for vigorous antiplasmodial activity while restraining potentially lethal immunopathologic damage to the host like cerebral malaria. Coinhibitory cell surface receptors are important modulators of immune activation. B and T lymphocyte attenuator (BTLA) (CD272) is a coinhibitory receptor expressed by most...
متن کاملThrombocytopenia in patients with melanoma receiving immune checkpoint inhibitor therapy
BACKGROUND Immune checkpoint inhibitors, including antibodies against programmed death 1 (PD-1) and cytotoxic T-lymphocyte antigen 4 (CTLA-4), are being used with increasing frequency for the treatment of cancer. Immune-related adverse events (irAEs) including colitis, dermatitis, and pneumonitis are well described, but less frequent events are now emerging with larger numbers of patients treat...
